Phenibut can be synthesized from the reaction between β-phenyl-GABA and hydrochloric acid. The synthesis process involves the following steps:


  1. Acylation of glycine with phenylacetyl chloride: In this step, glycine is reacted with phenylacetyl chloride in the presence of an organic base such as triethylamine to yield β-phenyl-GABA.
  2. Cyclization of β-phenyl-GABA: The β-phenyl-GABA is then reacted with phosphoryl chloride to form β-phenyl-γ-aminobutyryl chloride. This is then reacted with sodium hydroxide to form the final product, Phenibut.

The complete synthesis reaction can be represented as follows:

β-phenyl-GABA + PCl3 --> β-phenyl-γ-aminobutyryl chloride


β-phenyl-γ-aminobutyryl chloride + NaOH --> Phenibut
